Nature Methods Publication Reveals Deep Insights into Functional Biology through First-of-its-Kind Quantification of Tau Proteoforms
Nautilus Biotechnology published a peer-reviewed study in Nature Methods demonstrating that its Iterative Mapping approach quantified 130 tau proteoform groups at the intact, single-molecule level — a view no other available technology has been shown to produce.
Technically, the assay used 12 site-specific antibodies to resolve up to 768 possible proteoform groups, detected proteoforms present at just 0.1% of a sample with a median coefficient of variation below 5.5%, and showed tau modifications accumulate in ordered, non-random sequences tied to disease biology.
The same validated approach is being extended to α-synuclein in Parkinson's disease and to AKT1, Nautilus' first oncology assay expected to enter the Early Access Program in late 2026, with roughly 20 proteoform assays targeted by mid-2028.
For a development-stage company whose platform is not yet commercially available, this is a scientific credibility and pipeline-validation milestone rather than a near-term financial catalyst; the tau assay is available now through the Early Access Program.
